Sugar Determination in Dairy Products: Methodology

The sugar analysis is performed with MIC-8+Dialysis. The system is equipped with the Metrosep Carb 1 - 250 sugar column; a sodium hydroxide eluent and a pulsed amperometric detector are used. For the analysis of protein-containing samples a dialysis unit is used for the inline sample preparation. Unskimmed milk, yoghurt, baby food and dairy-industry intermediates (protein concentrates, milk powder, chocolate milk powder) are analyzed.

The patented stopped-flow method allows to achieve a practically complete dialysis. This means that after a dialysis time of 14 min the acceptor solution has the same analyte concentration as the sample (equilibrium dialysis). Read more about Metrohm Inline Dialysis as one variant of Metrohm Inline Sample Preparation (MISP).

The inline sample preparation procedure occurs in 4 steps

1. Rinsing of the sample preparation system

2.

Dialysis schemaDialysis of the sample (with flowing sample and stationary acceptor solution)

3.

Transfer schemaTransfer of the acceptor solution, which now contains the analyte, into the injection valve with flowing sample and acceptor stream

4.

Injection of the acceptor solution into the chromatography system
